چکیده

In the present study, we evaluate the protective effect of nitric oxide (NO) against the senescence of rice leaves promoted by hydrogen peroxide (H 2 O 2 ). Senescence of rice leaves was determined by decreases in protein content. H 2 O 2 treatment resulted in (1) increases in leaf H 2 O 2 content, (2) induction of leaf senescence, (3) increases in lipid peroxidation, (4) decreases in ascorbic acid (AsA) and reduced form glutathione (GSH) contents, and (5) increases in antioxidative enzyme activities (ascorbate peroxidase, glutathione reductase, and peroxidase). NO donors [N-tert-butyl-α-phenylnitrone (PBN), sodium nitroprusside, 3-morpholinosydonimine, and AsA + NaNO 2 ] were effective in reducing H 2 O 2 -induced leaf senescence. PBN prevented H 2 O 2 -increased H 2 O 2 content, H 2 O 2 -induced lipid peroxidation, H 2 O 2 -decreased AsA and GSH contents, and H 2 O 2 -increased antioxidative enzyme activities. The protective effect of PBN on H 2 O 2 -promoted senescence, H 2 O 2 -increased H 2 O 2 content and lipid peroxidation, H 2 O 2 decreased AsA and GSH contents, and H 2 O 2 -increased antioxidative enzyme activities was reversed by 2(4-carboxy-2-phenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide, a NO-specific scavenger, suggesting that the protective effect of PBN is attributable to the NO released. Reduction of H 2 O 2 -induced senescence by NO in rice leaves is most likely mediated through its ability to scavenge H 2 O 2 .

Arabidopsis nitric oxide synthase1 is targeted to mitochondria and protects against oxidative damage and dark-induced senescence.

The Arabidopsis thaliana protein nitric oxide synthase1 (NOS1) is needed for nitric oxide (NO) synthesis and signaling during defense responses, hormonal signaling, and flowering. The cellular localization of NOS1 was examined because it is predicted to be a mitochondrial protein. NOS1-green fluorescent protein fusions were localized by confocal microscopy to mitochondria in roots. Isolated mit...

Abscisic acid-induced hydrogen peroxide is required for anthocyanin accumulation in leaves of rice seedlings.

The role of hydrogen peroxide (H(2)O(2)) in abscisic acid (ABA)-induced anthocyanin accumulation in detached and intact leaves of rice seedlings was investigated. Treatment with ABA resulted in an accumulation of anthocyanins in detached rice leaves. Dimethylthiourea, a chemical trap for H(2)O(2), was observed to be effective in inhibiting ABA-induced accumulation of anthocyanins. Inhibitors of...

Protective effect of exogenous nitric oxide on alleviation of oxidative damage induced by high salinity in rice (Oryza sativa L.) seedlings

To find the protective role of exogenous nitric oxide (NO) on salinity-stressed rice seedlings, a CRD-based factorial experiment with three replications was conducted in Agronomy Laboratory of the Faculty of Agricultural Sciences, University of Guilan, in 2012. The experimental design consisted of healthy and vigorous seedlings of two rice cultivars, Khazar and Goohar, the last already known as...

Effect of oxidative stress caused by hydrogen peroxide on senescence of rice leaves

Lipid peroxidation is considered to be an important mechanism of leaf senescence. The peroxidation of lipids can be initiated by free radicals. H 2 O 2 itself is active oxygen species and can react with superoxide radicals to form more reactive hydroxyl radicals in the presence of trace amounts of Fe or Cu. Thus, it is of interest to investigate the relationship between lipid peroxidation and H...

Nitric oxide and protein S-nitrosylation are integral to hydrogen peroxide-induced leaf cell death in rice.

Nitric oxide (NO) is a key redox-active, small molecule involved in various aspects of plant growth and development. Here, we report the identification of an NO accumulation mutant, nitric oxide excess1 (noe1), in rice (Oryza sativa), the isolation of the corresponding gene, and the analysis of its role in NO-mediated leaf cell death. Map-based cloning revealed that NOE1 encoded a rice catalase...
